Physical and Chemical Properties | Bioactive Ilexsaponin A was isolated from the roots of Rabdosia rubescens and attenuates myocardial injury induced by ischemia-reperfusion via an anti-apoptotic pathway. Ilexsaponin A can reduce the size of myocardial infarction, reduce serum levels of LDH,AST and CK-MB, increase cell viability and inhibit apoptosis of hypoxia/reoxygenation cardiomyocytes. |
In vitro study | Ilexsaponin A significantly reduces proapoptotic proteins including caspase-3, cleaved caspase-3 and bax and increases anti-apoptotic protein bcl-2 in hypoxia/reoxygenation cardiomyocytes. Ilexsaponin A treatment increases the expression levels of p-Akt in hypoxia/reoxygenation cellular model and myocardial ischemia/reperfusion animal model. |
In vivo study | Ilexsaponin A (10 or 40 mg/kg) reduce the myocardial infarct size in a dose dependent manner in male Sprague-Dawley rats weighing 280-320 g. |

background
Ilex puplica is the dry root of Ilex puplica IlexpubescensHooketArm of Ilex puplica, a plant of the genus Ilex of the Ilex family. It grows in southern China and has the effects of clearing away heat and detoxification, promoting blood circulation and dredging veins, reducing swelling and relieving pain. In recent years, many studies on Ilex pupae have proved that Ilex pupae contains a large number of saponins. The herb contains a variety of chemical components. In addition to the main active ingredient triterpene saponins, it also includes flavonoids, lignans, simple phenols, coumarins and sugars. Studies have shown that triterpenoid saponins have obvious pharmacological effects and are the focus and hotspot of researchers in recent years. Moreover, with the deepening of the research topic of Ilex mausingii, some new triterpene saponin compounds have been extracted and isolated one after another.
detection method
ODS chromatographic column (4.6mm × 250mm,5 μm) and protective column were used, methanol-(volume fraction) 0.2% glacial acetic acid (67: 33) was used as mobile phase, evaporative light scattering detector, drift tube temperature was 95 ℃, carrier gas flow rate was 2.5L · min-1, column temperature was 30 ℃, flow rate was 1.0mL · min-1, the chromatographic conditions for determination of the content of paroxiferyl saponin A by high performance liquid chromatography, the results showed that the linear relationship between 1.92 and 9.60 μg was good (r = 0.9995), the average recovery rate was 99.85%,RSD was 1.04%. The contents of saponin A in Aikeqing capsule were:(10.89±0.04)mg · g-1,(11.23±0.03)mg · g-1,(10.46±0.06)mg · g-1 respectively.
Pharmacokinetics
In vitro cultured rat intestinal bacteria, Ilex pilosum saponin A is mainly metabolized by holly A; In vivo experiments, no prototype and metabolites were detected in urine, and prototype and metabolites were detected in feces. After oral administration of Ilex pilosum saponin A, the 89.85% is excreted from the body after metabolism by intestinal flora, of which 15.58% is excreted as prototype Ilex pilosum saponin A and 74.27% as metabolite holly A.
